| dc.description.resumo | A poluição atmosférica representa uma das principais ameaças à saúde pública e está associada a doenças cardiorrespiratórias. Embora alguns efeitos gestacionais da exposição ao material particulado fino (MP2,5) já tenham sido descritos, seus impactos sobre o sistema cardiovascular da prole, bem como o papel do treinamento físico nessa condição não são conhecidos. Este estudo teve como objetivo investigar os efeitos do treinamento físico em parâmetros morfofuncionais cardíacos, autonômicos e inflamatórios na prole de ambos os sexos de ratas Wistar expostas ao MP2,5 durante a gestação. As mães foram expostas ao MP 2,5 em um concentrador de partículas do 5o ao 21o dia de gestação. A prole foi distribuída em grupos de machos (M) e de fêmeas (F) (n=6-8 sexo/grupo): controles (CM, CF), expostos (EM, EF) e expostos treinados (TM, TF). O treinamento físico (8 semanas, 40-60% da velocidade máxima) foi iniciado após o desmame, seguido de avalições de morfometria e função cardíaca por ecocardiograma, da variabilidade da frequência cardíaca (VFC) por métodos lineares e não lineares e da inflamação sistêmica por ELISA. A exposição gestacional induziu a redução do diâmetro diastólico do ventrículo esquerdo (LVEDD;d), aumento da IL-6 e da razão IL-6/IL-10 em ambos os sexos. No grupo EM observou-se redução da massa do ventrículo esquerdo (VE) avaliada pelo eco e pelo peso úmido do tecido, da velocidade do anel mitral durante a sístole (S’) e durante o enchimento ventricular tardio (A’), acompanhada do aumento da relação enchimento precoce/tardio do VE (E/A). No grupo EF houve redução da variância do intervalo de pulso (Var IP). O treinamento físico aumentou a capacidade funcional, a fração de ejeção (FE), a Var IP e o componente vagal (HF), enquanto reduziu o TNF-alfa, a IL-6, a razão IL-6/IL-10 e a razão SD2/SD1 (desvio padrão instantâneo/longo prazo) em ambos os sexos. Especificamente no grupo TM, houve aumento da espessura do septo (IVS;d) e da parede posterior (PPVE;d) do VE, com redução do tempo de relaxamento isovolumétrico (TRIV) e do índice de performance miocárdico (IPM). No grupo TF, observou-se aumento do IVS;d, da S’ e da IL-10. Nas análises de correlação de Pearson, observou-se correlação negativa entre Var IP e IL-6/IL-10 (r = -0,62; p = 0,0014), bem como entre IL-6/IL-10 com MVE (r = -0,76; p = 0,0001) e com o IPM (r = -0,79; p = 0,0001), sugerindo interação entre disfunção autonômica, desequilíbrio inflamatório e comprometimento estrutural e funcional cardíaco. A exposição gestacional ao MP2,5 promoveu alterações morfofuncionais e inflamatórias persistentes, com maior suscetibilidade dos machos. O treinamento físico aeróbico atenuou essas disfunções, exercendo papel cardioprotetor e preventivo na prole. | por |
| dc.description.abstract | Air pollution poses one of the main threats to public health and is associated with cardiorespiratory diseases. Although some gestational effects of exposure to fine particulate matter (PM2.5) have already been described, its impacts on the cardiovascular system of offspring, as well as the role of exercise training in this context, remain unknown. This study aimed to investigate the effects of exercise training on cardiac, autonomic, and inflammatory morphofunctional parameters in the offspring of both sexes of Wistar rats exposed to PM2.5 during pregnancy. The mothers were exposed to PM2.5 in a particle concentrator from the 5th to the 21st day of gestation. The offspring were divided into male (M) and female (F) groups (n=6–8 per sex/group): controls (CM, CF), exposed (EM, EF), and exposed-trained (TM, TF). Exercise training (8 weeks, 40–60% of maximum speed) was initiated after weaning, followed by assessments of cardiac morphology and function by echocardiography, heart rate variability (HRV) using linear and nonlinear methods, and systemic inflammation by ELISA. Prenatal exposure led to a reduction in left ventricular end-diastolic diameter (LVEDD;d) and increased IL-6 and IL-6/IL-10 ratio in both sexes. In the EM group, a reduction was observed in left ventricular (LV) mass as assessed by echocardiography and tissue wet weight, as well as in mitral annular velocity during systole (S’) and during late ventricular filling (A’), accompanied by an increase in the LV early-to-late filling ratio (E/A). In the EF group, there was a reduction in pulse interval variance (Var IP). Exercise training increased functional capacity, ejection fraction (EF), Var IP, and the vagal component (HF), while reducing TNF-alpha, IL-6, the IL-6/IL-10 ratio, and the SD2/SD1 ratio (instantaneous/long-term standard deviation) in both sexes. Specifically in the TM group, there was an increase in the thickness of the septum (IVS;d) and the posterior wall (PPVE;d) of the LV, with a reduction in isovolumetric relaxation time (IVRT) and the myocardial performance index (MPI). In the TF group, an increase in IVS;d, S’, and IL-10 was observed. In Pearson correlation analyses, a negative correlation was observed between Var IP and IL-6/IL-10 (r = -0.62; p = 0.0014), as well as between IL-6/IL-10 and LV mass (r = -0.76; p = 0.0001) and with IPM (r = -0.79; p = 0.0001), suggesting an interaction between autonomic dysfunction, inflammatory imbalance, and structural and functional cardiac impairment. Prenatal exposure to PM2.5 caused persistent morphofunctional and inflammatory changes, with males showing greater susceptibility. Aerobic exercise training attenuated these dysfunctions, exerting a cardioprotective and preventive role in the offspring. | eng |
